摘要
目的:通过检测非酒精性脂肪肝形成过程中肝型脂肪酸结合蛋白的表达变化探讨其发病机制。方法:雄性大鼠60只,体重(180±10)g,随机分为饮食基础饲料的正常对照组和饮食高脂饲料(基础饲料87.8%、猪油10%、胆固醇2%、胆盐0.2%)的实验组;各组又随机分为2、4、8、12周4个时相组,用Western-blot方法进行L-FABP蛋白质的动态检测并探讨其发病机制。结果:L-FABP在高脂饮食后的第2周开始升高,与正常饮食组相比升高有显著性(1.31±0.02 vs0.67±0.03,P<0.01)。第8周时升至最高(1.67±0.07vs0.67±0.03,P<0.01),第12周有所降低,但与第8周相比没有统计学意义。结论:随着高脂饮食时间的延长,L-FABP的表达逐渐上升,一定程度上说明其和脂肪肝的进展有密切关系,有望成为脂肪肝治疗药物的靶点。
Objective-To investigate the mechanism of NAFLD by detecting the expression of L-FABP, NAFLD rat model development. Methods:Sixty male Wistar rats of (180±10)g weight were divided into two groups randomly:control group was fed with normal diet;Model group was fed with high fat diet (addition of 0. 1 lard oil,0. 02 cholesterol and 0. 002 cholate in mass fraction to normal diet) to establish NAFLD models. Then the two groups were sacrificed at the 2nd,4th,8th and 12th weeks,respectively. L-FABP was detected with Western-blot. Results:L-FABP expression was increased obviosely from the 2nd week comparied with control(1.31 ±0. 02 vs0. 67±0. 03, P〈0.01 ); L-FABP expression most obviosely at the 8th week(1.67±0. 07vs0.67± 0.03, P 〈0.01 ) and decresed a bit at 12th week, but no obviosely comparied with the 8th week. Conclusion:In the N AFI.D rat model, L-FABP expression is gradually increased, with some relationship between NAFI.D and L-FABP. L-FABP may become the target of medicine.
