摘要
目的观察黄连素对高脂膳食加小剂量链脲佐菌素(STZ)诱导的2型糖尿病(2-DM)大鼠胰岛素抵抗指数及肾周脂肪组织脂联素基因mRNA表达的影响。方法选用高脂饲料加小剂量STZ建立2-DM大鼠模型共40只,将2-DM大鼠随机分为模型组(水)和黄连素高、中、低剂量组(0.20、0.10、0.05g/kg),每组10只,另选10只大鼠为正常组,各组灌胃8周后,空腹取材,采用逆转录聚合酶链式反应检测肾周白色脂肪组织脂联素基因mRNA水平,放射免疫法测定空腹胰岛素并计算胰岛素抵抗指数(IRI)。结果正常对照组大鼠与2型糖尿病模型组大鼠脂联素基因表达、IRI间比较差异有统计学意义(P〈0.01);模型组与黄连素高、中剂量组大鼠脂联素基因表达、IRI间差异有统计学意义(P〈0.01)。结论黄连素可增加2-DM大鼠脂联素基因mRNA的表达,降低IRI。
Objective To explore the effect of Berberine decotion on adiponectin gene mRNA expression in white adipose tissue and insulin resistant index of 2-DM induced by hight-fat diat and streptozotocin (STZ). Methods To build rats model of type 2-DM induced by hight-fat diet and STZ. The rats with 2-DM were divided into four groups randomly:model groups and low, middle, hight doses of berberine groups with 10 in each group,another 10 rats were selected as normol group. After 8 weeks, FINS were detected by radioimmunoassay and was calculated insulin resistant index ( IRI), mRNA expression of adiponection gene was determinded by RT-PCR. Results There is great statistical difference between control group and 2-DM group about their gene expression of adiponectin and IRI( P 〈 0.01 ). So did the model group and berberine group with middle and high dosage. ( P 〈 0.01 ). Conclusion Berberine can increase mRNA expression of adiponection gene and decrease IRI in 2-DM rats.
