rhBMP-2 cDNA转染的NIH-3T3细胞对小鼠急性放射损伤的基因治疗作用与机理 被引量:4

摘要 探讨重组人骨形成蛋白-2(rhBMP-2)cDNA转染的NIH-3T3细胞对小鼠经6.5Gy,7.0Gyγ射线全身照射引起的放射损伤的基因治疗作用及其机理.方法:小鼠照射后1d和3d于腹腔内移植转染的细胞各1次,每次细胞量为1×107.观察动物的30d活存情况及几种造血指标.结果:治疗组的30d活存率、平均活存时间、CFU-GM集落形成数、骨髓细胞DNA含量、骨髓有核细胞数、脾结节形成数、脾体比及外周血白细胞数均较阴性对照组增加,差别非常显著(P<0.01).结论:rhBMP-2cDNA转染的NIH-3T3细胞对小鼠的放射损伤有治疗作用,其机理同它分泌rhBMP-2,诱导造血因子释放.改善造血微环境和促进造血功能恢复有关.
