摘要
目的:探讨肿瘤坏死因子(TNF)和化疗药物联合应用的作用机制。方法:用3AO卵巢癌细胞株作为靶细胞,体外培养,分单独用药及联合用药组,于用药后不同时间收获细胞,制成电镜标本,在透视电镜下观察单独应用TNF及TNF与化疗药物(阿霉素、更生霉素)联合用药对卵巢癌细胞超微结构的改变。结果:电镜显示,单独应用TNF,卵巢癌细胞无明显改变,联合用药后靶细胞改变出现时间早,程度重,主要在细胞质,表现为线粒体水肿、嵴消失,内质网增生、扩张,细胞空泡样变性多见,而细胞膜变化不明显。
Objective:To study the probable mechanism of treatment of ovarian cancer by combining TNF with chemotherapeutic drugs.Methods: Ovarian cancer cell line 3AO was used as the target cell and cultured in vitro. The cultural cells were divided into 3 groups, including 2 groups of using drugs(using TNF alone and using TNF combined with chemotherapeutic drugs) and control group. The cells were harvested after 6,12,24 and 48 h of culture to observe the ultrastructural changes of 3AO cells by using TNF alone and using TNF combined with chemotherapeutic drugs (adriamycin,actinomycin).Results: Under electronic microscope, it was found that injury to the target cell was unobvious by using TNF alone, but the injury to the target cell was serious and appeared early by using TNF combined with chemotherapeutic drugs. Cell change was in the cytoplasm, including swelling and crest disappearing of mitochondria, proliferation and enlargement of endoplasm reticulum, and increased intracytoplasmic vacuolar degeneration. The cytoplasmic membrane remained unchanged. Conclusion: Using TNF combined with chemotherapeutic drugs has the synergistic effect on ovarian cancer cell.
