摘要
以健康人外周血单个核细胞作为肿瘤杀伤细胞的前体细胞,MGc803胃癌细胞作为刺激细胞,在体外进行同种异体淋巴细胞和肿瘤细胞混合培养之后,单独加入重组白介素2和同时加入重组白介素2、抗CD3单克隆抗体,分别诱导出L肿瘤杀伤细胞和CD3肿瘤杀伤细胞.用噻唑蓝比色法在体外检测其对MGc803胃癌细胞的杀伤活性,并与淋巴因子激活杀伤细胞、CD3肿瘤杀伤细胞相比较结果显示,CD3肿瘤杀伤细胞的杀伤活性最高.在倒置显微镜下及透射电镜下观察CD3肿瘤杀伤细胞对MGc803胃癌细胞的杀伤过程,发现CD3肿瘤杀伤细胞首先接触、粘附靶细胞。
Tumor activated killer cells(TAK)were generated in human peripheral blood imononuclear cells by stimulating with human gastric carcinoma cell line MGc803 using allogenetic mixed lymphocyte tumor cell culture method. LTAK cells and CD3TAK cells were induced by MGc803 cells with rIL2, and MGc803 cells with rIL2,and antiCD3Ab,respectively.Testing in vitro cytotoxicity against MGc803 cells in MTT colorimetry CD3TAK cells gave the highest levels of cytotoxity compared with LAK cells and CD3AK cells.In vitro cytolysis process of MGc803 cells mediated by CD3TAK cells were observed using inverted microscopy and transmission electron microscopy.The results showed that CD3TAK cells attached the target and the target cells died by apoptosis and necrosis.
