Nm23-H1 is a metastasis suppressor gene involved many carcinomas, in addition, it correlated to cell proliferation, differentiation, cell signal transduction individual development and so on. However, the molecular mechanism by which nm23-H1 suppresses the metastasis is still unclear. Our previous studies in which Prof. Qinghua Zhou was the principal investigator demonstrated that transfection of exogenous nm23-H1 into the L9981 increased cell apoptosis and decreased metastatic ability. Furthermore, we found that the expression of β-Catenin, E-cadherin and TIMP-1 were significantly increased while the expression of Ras, MMP-2, CD44v6, and VEGFC dramatically decreased in L9981 cell line transfected with exogenous nm23-H1. We also found that G0/G1 phase cells markedly increased after transfection of exogenous nm23-H1. Here, for further confirm and discuss the correlation between tumor metastasis and nm23-H1, meanwhile as an extensive study and research, we compared the gene expression profile and biological behaviors of human large cell lung cancer cell line NL9980 by nm23-H1 gene silencing with that of negative control cells to comprehensively investigate nm23-H1-mediated changes in gene expression of NL9980 cells and further analyze how nm23-H1 as a key tumor suppressor gene regulate tumor metastasis/metastasis suppressor balance, which could lead to cascading response.
