摘要
目的观察穴位埋线和胰岛素联合应用对拟阿尔茨海默病(Alzheimer’s disease,AD)大鼠学习记忆的影响,并探讨其可能的治疗机制。方法大鼠海马CA1区微量注射冈田酸(Okadaic acid,OA)建立拟AD模型大鼠。肾腧、大椎穴位埋线、胰岛素侧脑室注射。用Morris水迷宫实验检测其学习记忆能力,电镜观察海马CA1区神经元超微结构变化,免疫组化方法观察胆碱乙酰转移酶(ChAT)的表达。结果与模型组比较,穴位埋线组、联合组的学习记忆能力明显改善(P<0.01),海马CA1区神经元凋亡现象明显改善,ChAT表达增加(P<0.05),联合组更为显著(P<0.05)。结论穴位埋线和胰岛素联合应用对拟AD大鼠学习记忆的改善作用强于单用穴位埋线,可能和改善海马CA1区神经元超微结构及胆碱能系统功能有关。
Objective To study the effects and its possible mechanism of Catgut implantation at acupoint and Insulin on the learning and memory abilities of Alzheimer' s Disease (AD) rats. Methods Okadaic acid(OA) was injected into hippocampal CA1 region of rats with microdosis repeatedly to establish Alzheimer' s disease - like model , and then insulin was injected into the lateral ventricle of cerebrum and catgut was implanted in Shenshu and Dazhui acupucture point. The learning and memory capacity of rats were assessed through Morris Water Maze behavioral test. Uhrastructure was observed by electron miscroscope and the expressions of ChAT apoptosis of neurons were examined by immunohistochemistry in hippoeampus CA1 region of the rats. Results Compared with model group, learning and memory capacity was improved obviously, and the expression of ChAT in hippocampal CA1 region was increased significantly in rats of catgut implantation at acupoint group and combination group, moreover, the effects were better in combination group. Conclusion Combination of Catgut implantation at acupoint and Insulin is more effective than only Catgut implantation at acupoint in Alzheimer' s disease rats, the mechanism might be related with improving uhrastructure in hippocampal CA1 region and function of central cholinergic system.
