摘要
目的:观察哮喘大鼠肺组织中p-Akt、p-p70S6K含量变化,探讨磷脂酰肌醇3-激酶(PI3K)信号转导途径在哮喘大鼠气道平滑肌细胞(ASMC)增殖中的作用。方法:SPF级健康雄性SD大鼠36只,随机分为对照组(C组)、哮喘组(A组)和wortmannin干预组(W组),每组12只。以卵蛋白(OVA)致敏和激发的方法制备大鼠慢性哮喘模型。观察各组大鼠气道炎症和细胞浸润情况;以图像分析软件测定气道壁厚度(Wat)及气道平滑肌层厚度(Wam);以Western blot法检测肺组织p-Akt及p-p70S6K表达情况。结果:①A组大鼠Wat、Wam大于C组(P<0.01);W组大鼠Wat、Wam小于A组(P<0.01),大于C组(P<0.01)。②A组p-Akt及p-p70S6K含量均高于C组(P<0.01);W组低于A组(P<0.01),但高于C组(P<0.01)。③p-Akt蛋白含量与Wam/Pbm呈显著正相关(r=0.779,P<0.01);p-Akt蛋白含量与p-p70S6K蛋白含量呈显著正相关(r=0.803,P<0.01)。结论:哮喘大鼠肺组织中PI3K/Akt/p70S6K途径激活,抑制该途径可减轻哮喘大鼠ASMC增殖。
Objective: To observe the changes of p-Akt, p-p70^s6k in lung tissues of asthma rat and to evaluate the role of phosphoinositide 3-kinase signal pathway in ASMCs proliferation in asthma. Methods: Thirty-six male Sprague-Dawley rats were randomly divided into three groups on average, including control group (group C), asthma group (group A) and wortmannin treated group (group W). In this experiment the rat model of chronic asthma was expioited, in which rats were sensitized with ovalbumin and Al (OH) 3' and repeatedly exposed to aerosolized ovalbumin. Airway remodeling was obtained by measurement and calculation of Wat and ram under microscope, p-Akt protein and p-p70^s6k protein in rat lung tissues were measured by western-blot. Results: ①Wat and Wam in group A were significantly increased than that of group C (P〈0.01); War and Wam in group W was decreased than that of group A (P〈0.01), but still higher than that of group C (P〈0.01). ②The leveI of p-Akt and p-p70^s6k protein in group A were both significantly higher than that of group C (P〈0.01). And the level of p-Akt and p-p70^s6k protein in group W were lower than that of group A (P〈0.01), but still higher than that of group C (P〈0.01). ③There was a significant positive correlation between p-Akt and Wam/Pbm. There was a significant positive correlation between p-Akt and p-p7^s6k. Conclusion: The expression of p-Akt and p- p70^s6k increased in lung tissues of asthmatic rat model and wortmannin can reduce ASMCs proliferation, which implicates that PI3K signal transduction pathway plays an important role in the ASMCs proliferation of asthma.
