摘要
目的观察外源性骨髓间充质干细胞在活体兔椎间盘微环境中存活状态。方法选取健康新西兰大白兔24只,体外培养经基因转染红色荧光蛋白标记的兔骨髓间充质干细胞,待细胞活性状态最佳时,利用显微注射方法将其注入兔腰椎间盘,分别于术后1、7、14、30、60 d手术取出椎间盘,制备病理学切片,在荧光显微镜下观察外源性骨髓间充质干细胞是否散发出红色的荧光及荧光细胞数量。结果术后1、7、14、306、0 d椎间盘切片在荧光显微镜下均可观察到荧光,术后14 d细胞所散发出的荧光较术后1 d及7 d有所减弱,术后60 d红色荧光无明显变化。术后306、0 d荧光细胞数量较1-14 d有所增加(F=15.171,P〈0.01)。结论手术后60 d外源性骨髓间充质干细胞在活体兔椎间盘内仍处于存活状态。
Objective To observe the survival condition of exogenous mesenchymal stem cells (MSC) in microenvironment of intervertebral disc (IVD) of rabbit. Methods Twenty-four healthy New Zealand white rabbits were selected, and the cells were micro injected into rabbit IVD when the MSCs were at the best active state. The IVD was removed on day 1, 7, 14, 30 and 60 after operation, and a pathological section done. To make sure if red fluorescence was erupted by MSC and the number of fluorescyte, an observation was done with fluorescence microscope. Results The red fluorescence could be seen on all IVD sections, the fluorescence on 14th day after injection was weaker than that on 1st and 7th day, and obvious red fluorescence seen on 60th day, the cells were more diffused distribution. The number of fluorescytes was more on 30th and 60th day than that on day 1-14 (F=15. 171,P〈0.01). Conclusion Exogenous MSCs can still survive in living rabbit IVD 60 days after surgery.
