金属螯合亲和层析法对人源宋内痢疾杆菌脂多糖抗独特型抗体重链可变区蛋白的纯化 被引量:4

摘要 在重组人宋内痢疾杆菌脂多糖抗独特型抗体重链可变区蛋白的表达质粒(pQEHv)的氨基端插入6个组氨酸,使其对金属螯合层析介质能产生特异性吸附,可用金属螯合亲和层析法进行分离纯化。本研究采用NiNTA螯合层析介质,以咪唑洗脱表达的蛋白。经ELISA检测,纯化的蛋白可特异地与抗宋内痢疾杆菌脂多糖的鼠mAb5B12发生反应。 Recombinant human Hv(heavy chain variable region of anti idiotypic antibody against antibody of LPS of Shigella sonnei(pQEHv) with a hexahistidine tail attacked to the N terminal of Hv could be easily purified by immobilized metal ion affinity chromatography (IMAC). Immobilized metal ion (Nickel) and elution solution(inidazole) were used in IMAC. The purified protein can react specially with 5B12 monoclonal antibody(anti LPS of Shigella sonnei)in ELISA.
