摘要
目的探讨经胆道灌注化疗对肝内胆管癌大鼠模型肝脏和胆管上皮的损伤以及对肿瘤组织的影响,评价灌注化疗的作用。方法选择自制的大鼠肝内胆管癌动物模型,经胆总管灌注化疗药物,观察药物对肝脏和胆管上皮的损伤情况,利用Western blot检测并对比肿瘤组织内Bcl-2、c-Met在灌注前后的表达情况。结果胆管化疗药物灌注可造成肝功能和胆管上皮的损伤,但该损伤动物可以耐受并在短期内恢复,Bcl-2、c-Met在灌注后1w对比灌注前表达程度有明显的下降,其中c-Met表达率的下降与对照组相比有统计学意义(P<0.01)。结论经胆管灌注并保留化疗药物治疗肝内胆管恶性肿瘤会造成肝脏和胆管上皮的可逆损伤,能够抑制肿瘤细胞的增生,诱导其凋亡。
Objective To survey the effect of transbiliary chemotherapeutic infusion on hepatoeytes, bile duct cells and tumor cells, assess its effect in cholangiocarcinoma model of rats. Methods Transbiliary chemotherapeutic infusion was performed on intrahepatic cholangiocarcinoma model of rats, tumor tissues, livers and plasma were acquired by operation after one and two weeks. The trauma in livers and bile duct epithelial were observed and the changes of some protein with Western blot were investigated, including Bcl-2 and c-Met. Results Like previous conclusion from normal rats, the damage to the liver and bile duct epithelial also existed, and could self-repair. Apparent apoptosis was found in epithelial cells in the bile duct, and the expression level of Bcl-2 and c-Met after infusion was lower than that before. Conclusion The combination of dose and time of drug maintenance in bile duct are also suitable for the model of rats. The way of transbiliary chemotherapeutic infusion suppresses the growth of tumor mass by inducing apoptosis.
