cAMP反应元件结合蛋白在2型糖尿病大鼠肝脏的表达与意义

Expression and its significance of cAMP-responsive element binding protein in liver of rats with type 2 diabetes mellitus

摘要 目的探讨cAMP反应元件结合蛋白(CREB)及磷酸化的CREB(p-CREB)在2型糖尿病(T2DM)大鼠肝脏的表达及其意义。方法健康雄性Wistar大鼠随机分为正常对照(NC)组和T2DM组。以高脂高糖喂养加小剂量STZ腹腔注射法制备T2DM大鼠模型。免疫组织化学和Western印迹法检测两组大鼠肝组织CREB及p-CREB蛋白的表达。结果与正常大鼠相比,T2DM大鼠肝脏p-CREB表达明显增高(P<0.01),血糖、血脂、肝TG、肝TC均与p-CREB相关;多因素逐步回归显示p-CREB与血糖独立相关。结论 T2DM大鼠肝脏p-CREB表达明显增强,CREB活性变化可能与糖脂代谢紊乱密切相关。 Objective To explore the expression of cAMP-responsive element binding protein (CREB) and phospho-CREB in liver of rats with type 2 diabetes mellitus. Methods Normal 6-week-old male Wistar rats were randomly divided into two groups: normal group (NC) and type 2 diabetic group (T2DM). Type 2 diabetic rat model was induced by high fat and high glucose-feeding and whereafter by plus low-dose STZ(30mg/kg) injection. The expression of CREB and phospho-CREB was assayed by immunohistochemistry and Western blot. Results The level of p-CREB in liver was higher in T2DM group than in NC group (P〈0. 01) and was independently correlated with blood glucose. Conclusions The expression of p-CREB is increased in liver of type 2 diabetic rats. It suggests that CREB may be highly correlated with abnormal hepatic lipid and glucose metabolism in T2DM.
