乳腺癌患者血清与肿瘤组织基质细胞衍化因子-1表达及其临床意义的研究 被引量:1

Expression of stromal cell derived factor-1 in sera and tumor tissues of patients with breast cancer and its clinical significance

摘要 目的:探讨基质细胞衍化因子-1(stromal cell-derived factor-1,SDF-1)在乳腺癌患者的血浆、肿瘤组织及淋巴结中表达水平及其与淋巴结转移状况之间的关系。方法:酶联免疫吸附试验(ELISA)检测患者血浆中的SDF-1蛋白水平;逆转录-聚合酶链反应(RT-PCR)半定量检测乳腺正常组织、良性肿瘤组织及乳腺癌组织中SDF-1mRNA水平;免疫组化法(IHC)检测乳腺正常组织、良性和恶性肿瘤组织及淋巴结中SDF-1蛋白的表达水平及分布情况。结果:良性与恶性肿瘤患者血浆中SDF-1蛋白表达为(39.98±10.38)和(56.21±10.37)pg/mL,P<0.01。乳腺癌组织SDF-1mRNA表达水平为0.596±0.257,较正常组织(0.291±0.032)和良性肿瘤组织(0.32±0.071)显著增加。乳腺癌组织中,有淋巴结转移者的SDF-1mRNA的表达水平(0.729±0.265)较无淋巴结转移者(0.419±0.104)有显著增加,P=0.034。IHC法显示,SDF-1蛋白在正常和良性肿瘤组织中呈现低水平表达,在乳腺癌组织中呈现高水平表达。有癌细胞转移的淋巴结其SDF-1蛋白水平表达较无癌细胞转移的淋巴结增加,P=0.015。结论:临床有望通过检测易于获得的血浆标本SDF-1蛋白水平预测淋巴结转移状况。 OBJECTIVE: To investigate the relationship between the levels of SDF-1 expression in blood plasma, tumor tissues and lymph nodes in patients with breast cancer and the state of lymph node metastasis. METHODS: The levels of SDF 1 protein in blood plasma were examined by enzyme linked immunosor bent assay (ELISA). The SDF-1 mRNA expressions in tumor tis sues and normal tissues were examined by semi-quantitative reverse transcription polymerase chain reaction(RT-PCR) technique. The expressions and distributions of SDF-1 protein in tumor tissues, normal tissues and lymph node tissues were studied by immunohistochemical staining (IHC). RESULTS: The levels of SDF 1 proteins detected in blood plasma of the patients with breast benign tumors or malignant tumors were (39.98 ±10.38) pg/mL or (56.21± 10.37) pg/mL, P〈0. 01. The level of SDF 1 mRNA expression was 0. 596±0. 257 in the tissue of breast cancer, which was significantly higher than those in the normal tissue(0. 291 ± 0. 032) and benign tumor tissue (0. 32± 0. 071). The breast cancer group with lymph node metastasis showed higher SDF1 mRNA expression (0. 729 ± 0. 265) than the cancer group without lymph node metastasis (0. 419 ± 0. 104), P = 0. 034. The immunohistochemistry methods showed that SDF-1 protein had low expressions in normal tissues and benign tissues, but had a high expression in malignant tumor tissues. Tumor tissues with metastasis showed a higher level of expression than tumor tissues without metastasis, P= 0. 015. CONCLUSION: It may be possible to predict the state of lymph node metastasis through testing the SDF-1 levels of the blood samples in the breast cancer patients in the future.
