蛋白激酶Cδ对人原发性肝癌细胞增殖的影响

摘要 目的初步探讨PKCδ对人原发性肝癌细胞增殖的影响.方法体外培养SMMC-7721细胞,脂质体介导基因体外转导,通过细胞计数、细胞形态学观察、MTT法、流式细胞仪检测技术等方法研究PKCδ对人原发性肝癌细胞的影响.结果细胞计数的生长曲线示转导入PKCδ的SMMC-7721细胞数目明显减少;MTT法显示转导入PKCδ的SMMC-7721细胞的OD值减小,活细胞数目显著减少(P<0.01);细胞形态学观察转导入PKCδ的SMMC-7721细胞脱落,连接疏松,失去光泽;流式细胞仪检测到加入PKCδ的SMMC-7721细胞凋亡率增加.结论PKCδ有抑制SMMC-7721细胞增殖,促进凋亡的作用. Objective To explore the effect of cell. Methods SMMC-7721 was cultured in vitro, PKCδon the grow the of human hepatoma carcinoma gene of PKCδ was transducted by liposomes, we reseached the effect of PKCδ on SMMC-7721 through cell counting, cell morphology observering, the method of MTT, the technique of flow cytometry. Results The cell growth curve displayed that the number of SMMC-7721 with PKCδ gradully discreased, the method of MTT displayed that the OD value of SMMC-7721 with PKC 8 dercreased, the number of living cell remarkably decreased (P 〈 0.01) , cell morphology displayed that cell abscisic, link loosen, lose glare; and the technique of flow cytometry displayed that SMMC-7721 apotosis rate increased. Conclusion PKCδ can inhibit the growth of SMMC-7721 and promote its apoptosis.
