摘要
目的以大鼠单纯胫骨骨折后加入骨粉复合物为模型,观察乙二胺四乙酸(EDTA)-2Na脱钙和混合酸脱钙后免疫组织化学检测的效果。方法将骨粉复合物放置于大鼠左侧胫骨的骨块缺损中建立人工骨成骨模型,将模型分为两组(每组标本10例),实验组用EDTA-2Na脱钙,对照组采取混合酸脱钙后分别观察两组的免疫组织化学检测效果。结果EDTA脱钙组骨小梁结构保存完整,阳性表达较多。混合酸脱钙后,骨结构破坏较多,抗原破坏较严重,阳性表达明显减少。结论EDTA脱钙优于传统的脱钙方法,更适合于骨组织脱钙和免疫组织学化制片。
Objective To investigate the antigenicity of osseous tissue influenced by Immunohistochemical staining method under different decalcification conditions. Methods Artificial bone sample was decalcificated under different conditions: The decalcification technique of EDTA-2Na and The decalcification technique of mixed acid. Results The number of positive cells was much greater in EDTA group than those in the groups processed with the mixed acid solutions. Conclusion The decalcification technique of EDTA-2Na is better than that of mixed acid.
