摘要
目的:探讨益气活血中药抑制血管平滑肌细胞(VSM)增殖的分子机制。方法:血管平滑肌细胞被培养在四种培养基中:常规培养基作为对照组,氧化型低密度脂蛋白组,以及在氧化型低密度脂蛋白组基础上的高浓度和低浓度两种兔载药血清培养基组。完成培养及干预后,检测PGC-1、NRF-1(核呼吸因子)和m tTFA(线粒体转录因子)蛋白表达,线粒体功能改变(COX酶活性)及细胞活性。结果:与对照组相比,氧化型低密度脂蛋白组PGC-1、NRF-1、m tTFA蛋白表达、线粒体COX(细胞色素C氧化酶)酶活性及细胞增殖活性均增加(P<0.05)。与氧化型低密度脂蛋白组相比,载药血清组PGC-1、NRF、m tTFA蛋白表达及COX酶活性降低(P<0.05);细胞增殖受到抑制(P<0.05),这种抑制效率呈现药物浓度依赖性。结论:益气活血汤药在人血管平滑肌细胞增生过程中通过改变线粒体功能影响细胞的生长状态。
Objective : To investigate the molecular biological mechanism of decoction with replenishing Qi and activating blood in inhibiting the proliferation of smooth muscle cell. Methods : Smooth muscle cells were cultured in four conditions , which were determined according to four kinds of serum culture - medium . They were the controls, the ox - LDL medium group,high -dose drug group and low -dose drug group. Then,PGC -1 ,NRF -1 and mtTFA were detected by western blot, and the functions of mitochondria and activity of cell were investigated. Results : Comparing to the control, the PGC - 1, NRF,mtTFA proteins of VSMC in ox -LDL group presented a higher levels, COX activity and proliferative index also increased( P 〈0.05). After the drug -containing serum were added into ox -LDL medium, the PGC - 1, NRF, mtTFA proteins of SMC, COX activity and proliferative index decreased significantly( P 〈 0.05 ), These changes showed a dose dependent. Conclusion : Decoction with replenishing qi and activating blood may affect the proliferative conditions of vescular smooth muscle cells via interfering the functions of mitochondria.
