摘要
目的通过观察卵清蛋白(OVA)诱导的变应性哮喘小鼠脾脏巨噬细胞中TLR2和TLR4 mRNA的表达变化,探讨复方丹参注射液(Complex Salvia Miltiorrhiza Injection,SA)对其作用的机制。方法将30只雄性BALB/c小鼠随机分为对照组、模型组和SA组,每组各10只。各组按要求处理于20天后取脾脏,贴壁法分离脾巨噬细胞,通过实时荧光定量聚合酶链反应(RT-PCR),检测TLR2和TLR4 mRNA以及GAPDH cDNA的荧光域值循环数,观察SA对TLR2和TLR4 mRNA水平表达的影响。结果与对照组相比,模型组TLR2 mRNA表达明显下调,差异有统计学意义(P<0.05),而TLR4 mRNA表达无变化(P>0.05);模型组与SA组比较,SA组TLR2和TLR4 mRNA表达均上调,且有统计学差异(P<0.05)。结论SA能使变应性哮喘模型小鼠脾巨噬细胞中TLR2和TLR4 mRNA水平表达上调,为中药在哮喘治疗中的应用提供了科学依据。
Objective To investigate the expression of TLR4 and TLR2 mRNA and the effect of SA on them in splenic macrophages of allergic asthma mice. Methods 30 BALB/c mice were divided into 3 groups : control group, model group and complex salvia miltiorrhiza injection (SA) group, with 10 mice in each group. After 20 days treatment, the spleen of three groups were sampled and adherent method was used to separate splenic macrophages, saline. Real-time quantitative RT-PCR was performed to evaluate mRNA production of GAPDH, TLR4 and TLR2 level of three groups. The influence of SA was evaluated by the expressions of TLR4 and TLR2 mRNA. Results Compared with the control group, the expression of TLR2 mRNA in the model group was downregulatod, there was difference (P 〈 0. 05 ), but there was no change in TLR4 mRNA between the two groups ( P 〉 0. 05 ). Compared with the model with SA group, the ex- pression of TLR2 and TLR4 mRNA in the splenic macrophages of SA group was upregulatod, and the difference was significant ( P 〈 0. 05). Conclusion SA could upreglatod the expression of TLR4 and TLR2, which provide scientific foundation for clinical practice.
