摘要
目的探讨高密度脂蛋白(high-density lipoprotein,HDL)预处理对缺血再灌注损伤(ische-mia-reperfusion injury,IRI)大鼠肺组织的影响。方法将24只SD大鼠随机分成3组:假手术组(SO)、缺血再灌注损伤组(I/R组)和缺血再灌注高密度脂蛋白预处理组(H/IPC组)。建立大鼠肺脏缺血再灌注模型,并检测各组PH、PaO2、PaCO2、肺湿/干重比值(wet-to-dry weight ratio,W/D)、支气管肺泡灌洗液(bronchial alveolar lavage fluid,BALF)中白细胞(White blood cell,WBC)计数、测定髓过氧化酶(myeloper-oxidase,MPO)水平,并观察肺组织病理变化。结果HDL改善大鼠换气功能,升高PaO2和降低PaCO2;②HDL降低肺组织湿/干重比值(W/D)和髓过氧化酶(MPO)的活性(P<0.05),降低肺泡灌洗液中WBC数量;③病理形态学显示,HDL预处理组大鼠在肺水肿及肺组织内中性粒细胞浸润的程度均低于I/R所致损伤组。结论经90min缺血,180min再灌后,HDL预处理能减轻I/R所致PaO2的降低、血管通透性增高,中性粒细胞浸润,减轻I/R大鼠肺组织损伤。
Objective To investigate the effects of HDL preconditioning on ischemia-reperfusion lung injury in rats. Methods Twenty-four SD rats were divided randomly into three groups : sham operation ( SO), ischemia-reperfusion groups ( I/R ), HDL/ ischemic-preeonditioning groups ( H/IPC ). The isehemia-reperfusion model was established, PaO2 、pH、PaCO2 in arterial , White blood cell (WBC)in the bronchial alveolar lavage fluid(BALF) samples , wet-to-dry weight ratio ( W/D ), myeloperoxidase ( MPO ) activity were measured. And Morphological changes of lung tissues were observed under light microscope. Results HDL-treated mice exhibited a higher PaO2, and pH, but a lower PaCO2 than HDL-untreated ones (P 〈0.01 ). WBC number in BALF was significantly attenuated in HDL-treated mice (P 〈 0.01 ). HDL treatment also resulted in a significant protection of lung tissues against I/R-induced acute lung injury via decreasing W/D ratio, MPO activity ( P 〈 0.05 ). It is revealed that HDL treatment resulted in significantly lower scores of acute lung injury induced by/JR, with reduced hemorrhage,intra-alveolar edema and neutrophilic infiltration. It is suggested that HDL plays a protective role in lung injury in mice.
