摘要
目的:初步探讨RUNX3基因在鼻咽癌中的失活机制。方法:采用甲基化特异性聚合酶链反应(MSP)法对50例鼻咽癌和20例鼻咽黏膜慢性炎组织中RUNX3基因启动子区CpG岛的甲基化水平进行检测。结果:在1例鼻咽癌组织中检测到RUNX3基因启动子区CpG岛甲基化,所有黏膜慢性炎组织中未检测到甲基化改变,二者相比差异无统计学意义(P=1.0000)。结论:RUNX3基因启动子CpG岛的甲基化可能不是RUNX3基因在鼻咽癌中转录失活的主要机制。
Objective:To investigate the inactivation mechanism of gene RUNX3 in nasopharyngeal carcinoma(NPC) preliminary. Methods. The methylation status of CpG island in RUNX3 gene promoter region in 50 nasopharyngeal carcinoma and 20 chronic nasopharyngitis tissues was determined by methylation-specific polymerase chain reaction(MSP) technique. Results. Aberrant methylation of CpG island in RUNX3 gene promoter region has been detected in only 1 of 50 nasopharyngeal carcinoma tissues, and none in chronic nasopharyngitis ones, and there is no significant difference( P = 1. 000 0). Conclusion. The methylation of CpG island in promoter region may not be the main mechanism of inactivation of RUNX3 in NPC.
