摘要
【目的】研究海马神经元在中度液压冲击损伤后PAFR、COX-2和2GluR2基因表达时序性变化的相关性。【方法】原代培养大鼠海马神经元,复制改良Scott′s中度液压冲击神经元损伤模型,采用免疫荧光双标技术鉴定神经元纯度,应用RT-PCR技术对PAFR mRNA、COX-2 mRNA和GluR2 mRNA的表达进行定量研究。【结果】对照组COX-2 mRNA呈低表达,损伤后表达逐渐增高,12 h达最高值,损伤后12 h组与对照组相比差别有显著性(P<0.05);对照组GluR 2mRNA呈高表达,损伤后表达逐渐减弱,8 h达最低值,损伤后8 h、12 h组与对照组相比差别有显著性(P<0.05);对照组PAFR mRNA呈高表达,损伤后表达逐渐减弱,8 h达最低值,损伤后8 h、12 h组与对照组相比差别有显著性(P<0.05)。【结论】正常时COX-2 mRNA在体外培养神经元中仅有少量表达,损伤后4 h表达增加,12 h达最高值,48 h时仍高于正常。正常时GluR2 mRNA在体外培养神经元有较多表达,损伤后4 h表达下降,8 h达最低值,48 h时仍低于正常。正常时PAFR mRNA在体外培养神经元中有较多表达,损伤后4 h表达下降,8 h达最低,48 h时仍低于正常。PAFR mRNA、COX-2 mRNA和GluR2 mRNA在体外海马神经元中度液压冲击损伤中表达的时序性变化特点基本一致,三者关系密切,共同参与脑损伤病理生理过程。
[Objective] To study the expressions of platelet activating factor receptor (PAFR), cyclooxygenase-2 (COX-2) and glutamate receptor 2 (GluR2) in the primary cultured hippocampal neurons of rats following fluid percussion injury in vitro, and to observe the temporal patters and relationship among their expressions following injury. [ Methods] To culture hippoeampal neurons of rats, and reprove Scott's model of themorphological changes of hippocampus neurons after fluid percussion injury, the localization of PAFR mRNA, COX-2 mRNA and GluR 2mRNA in cultured hippocampus neurons were studied with double immunofluorescenece staining. [Results] After injury, the expression of COX-2mRNA increased, which was highest at 12 h, and remained higher than control group ( P 〈 0.05) ; the expression of GluR2 mRNA decreased, which was lowest at 8 h, and remained lower than control group ( P 〈 0.05) ; the expression of PAFR decreased, which was lowest at 8 h, and remained lower than control group ( P 〈 0.05). [ Conclusions] After fluid percussion injury for 4 h, the expression of COX-2mRNA in cultured hippocampus neurons increased progressively, and reached the peak level after 12 h. It was mole than the normal after hippoeampus neurons injury for 48 h. COX-2mRNA was located in eeU membrane, cytoplasm, neurite and dendrite of hippoeampus neurons.After fluid percussion injury for 4 h, the expression of GluR2 mRNA in cultured hippocampus neurons decreased progressively, and reached thelowest level after 8 h. It was less than the normal until hippoeampus neurons injury for 48 h. GluR2 mRNA was mainly located in neurite, dendrite and the origin of neurite of cytoplasm hippocampus neurons. After fluid percussion injury for 4 h, the expression of PAFR mRNA in cultured hippocampus neurons decreased progressively, and then reached the lowest level after 8 h. It was less than the normal until hippocampus neurons injmy for 48 h. PAFR mRNA was mainly located in cell membrane, neurite and dendrite of hippocampus
