5种配方对草履虫生长及种群密度的影响 被引量:6

摘要 [目的]筛选适宜草履虫生长及繁殖的最佳营养配方及环境条件。[方法]采用5种营养配方,即配方Ⅰ-稻草煮剂培养液、配方Ⅱ-菠菜叶+稻草煮剂培养液、配方Ⅲ-稻草煮剂+牛奶液培养液、配方Ⅳ-0.1%蜂蜜培养液和配方Ⅳ-小麦粒培养液培养草履虫,并对草履虫生长发育和种群密度及理化指标进行比较。[结果]培养时间最短的是配方Ⅲ,仅用2d就可得体长为90~210μm,体宽为加~30脚,每毫升水样含1000~3000个草履虫,纯度也很高,能够用于纯培养,用该配方培养草履虫效果最佳。草履虫生长的最适温度是25℃,最适繁殖温度在10~30℃。pH值在5.0~10.0的范围内,草履虫的生长状况都较好,但最适合于在中性略偏酸的环境中生长。[结论]该研究为草履虫的生产应用奠定了基础。 [ Objective ] The aim was to screen the optimal nutrition formula and environmental condition suitable for growth and propagation of paramecium. [ Method] 5 kinds of nutrition fornadas of formula Ⅰ-culture medium of boiling straw, formula Ⅱleaves of spinach + culture medium of boiling straw, formula Ⅲ-culture medium of boiling shaw + culture medium of milk, formula Ⅳ-0. 1%-culture medium of honey, fommula Ⅴ-culmre medium of wheat were adopted to culture paramecium. The growth, population density and physical and chemical indices of paramecium were compared. [ Result ] Formula Ⅲ had shortest culture time. Paramecium could be acquired after 2 d culture, whose body length was 90- 210 μn and body width was 20- 30 μn, and there were 1 000- 3 000 paramecium in every lml water sample, the purity of paramecium was high and it could be used for pure culture. The effect of using the formula for culturing paramecium was best. The optinmm temperature for paramecium culture was 25 ℃ and optimum propagation temperature was 10- 30 ℃. The growth condition of paramecium was better when the pH value was in the range of 5.0- 10.0, but the optimum growth environment was neutral or near to alkalescence. [Conclusion] The research laid the foundation for production applieation of paramecium.
