摘要
研究了培养基组分对hCG核酸疫苗质粒拷贝数的影响。结果表明,复合培养基比合成培养基更利于hCG核酸疫苗质粒DNA的生产。通过均匀设计实验确定,当培养基中含有胰蛋白胨10g·L-1、酵母浸出粉8g·L-1、NaCl10g·L-1、甘油10g·L-1、KH2PO40.37g·L-1、K2HPO42.01g·L-1时,单位菌体质粒含量可达6.68mg·g-1;在此基础上添加乙酸2.0g·L-1、谷氨酰胺1.2g·L-1、甘氨酸1.0g·L-1、天门冬氨酸0.4g·L-1,可使质粒拷贝数进一步扩增,单位菌体质粒含量可达9.32mg·g-1,为LB培养基的5.12倍。
The effect of medium composition on plasmid amplification of hCG DNA vaccine was studied. The result showed that complex medium was better than defined medium for the production of plasmid DNA. The method of uniform design was applied to the design of culture medium. It was found that specific plasmid DNA yield'amounted up to 6.68 mg·g^-1 as the medium containing tryptone 10 g·g^-1, yeast extract 8 g·g^-1, NaCl 10 g·g^-1, glycerol 10 g·g^-1, KH2PO4 0.37 g·g^-1, K2HPO4 2.01 g·g^-1. Based on these, plasmid amplification increased dramatically through adding acetic acid 2.0 g·g^-1 ,Gln 1.2 g·g^-1 , Gly 1.0 g·g^-1 , Asp 0.4 g·g^-1. The final specific plasmid DNA yield reached 9.32 mg·g^-1 in the optimized medium, which was 5.12 times that in LB medium.
