摘要
为了克服传统切片染色中的切片污染和皱褶问题,尝试利用透射电镜制样中的标本块染方法.使用传统染色液在组织处理过程中对标本块进行染色,与传统的片染方法相比,可以简化制样步骤、减少切片污染和皱褶,但只有在样品浅层一定厚度范围内的切片才可以获得满意的观察效果.
En Bloc staining used in specimen preparation for transmission electron microscope observation was tried to reduce contamination and wrinkles of ultrathin sections occurring during standard routine specimen staining for transmission electron microscope observation. Compared with the routine method, En Bloc staining could simplify the process of preparation and reduce contamination of ultra thin sections. But satisfactory observation effect could only be achieved in those sections from superficial layer of specimen.
