摘要
目的:研究不同浓度重铬酸钾(K2Cr2O7)对人肺腺癌A549细胞的增殖抑制效应以及对O6-甲基鸟嘌呤-DNA甲基转移酶(hMGMT)蛋白表达的影响。方法:用四甲基偶氮唑蓝(MTT)比色法测定9种不同浓度K2Cr2O7(0.000、0.010、0.040、0.160、0.312、0.625、1.250、2.500、5.000μmol/L)对A549细胞的增殖抑制率;用损伤修复试验观察在不同浓度K2Cr2O7作用下人肺腺癌A549细胞的划痕修复情况;以β-actin为参照,应用Western blotting技术检测不同浓度K2Cr2O7对人肺腺癌A549细胞hMGMT表达的影响。结果:MTT比色法检测显示,大于0.625μmol/L浓度组K2Cr2O7可抑制细胞活性,与空白对照组比较,差异有统计学意义(P<0.05)。损伤修复试验显示,与空白对照组比较,0.625、1.250、2.500和5.000μmol/L的K2Cr2O7抑制细胞损伤修复,而0.312、0.160、0.040μmol/L的K2Cr2O7促进细胞损伤修复。Western blotting显示,0.312、0.160、0.040μmol/L的K2Cr2O7对hMGMT蛋白的表达具有促进作用,0.625、1.250、2.500和5.000μmol/L的K2Cr2O7对hMGMT蛋白的表达具有抑制作用。结论:低浓度的K2Cr2O7对人肺腺癌A549细胞的增殖、损伤修复和hMGMT蛋白的表达呈现低剂量兴奋效应;高浓度K2Cr2O7对hMGMT蛋白的表达则呈现抑制作用。
Aim:To investigate the effects of potassium dichromat on human lung adenoearcinoma A549 cell line in proliferation and hMGMT protein expression. Methods:Nine different concentrations of Potassium dichromat were administrated with methyl thiazolyl tetrazolium (MTT) assay to measure the inhibiting rates of cell growth. Wound-healing assay was applied to observe the condition of wound-healing. Western blotting was applied to determine the expression level of hMGMT protein. ReSults: The MTT assay showd that 0. 625 μmol/L, 1. 250 μmol/L, 2. 500 μmol/L and 5. 000 μmol/L K2Cr207 could restrain cell growth. Compared with the control group,the difference of cellular multiplication was different (P 〈 0.05). Wound-healing assay showed K2Cr207 can suppress cell repair with concentration of 0. 625μmol/L, 1. 250 μmol/L, 2. 500 μmol/L and 5. 000 μmol/L, and promote cell repair with concentration of 0. 312 Ixmol/L, 0. 160 μmol/L and 0. 040 μmol/L. Western blotting showed K2 Cr207 can promote the expression of hMGMT protein with concentration of 0. 312 μmol/L, 0. 160 μmol/L and 0. 040 μmol/L, and suppress the expression of hMGMT protein with concentration of 0. 625 μmol/L, 1. 250 μmol/L , 2. 500 μmol/L and 5. 000 μmol/L. Gonlusion: Low concentration of K2Cr207 showed hormisis phenomena of cellular multiplication,Wound-healing and hMGMT protein expression in human lung adenocarcinoma A549. Meanwhile, high concentration of K2 Cr207 could inhibit cellular multiplication, Wound-healing and hMGMT protein expression in human lung adenocarcinoma A549.
