摘要
应用蛋白质组学技术比较家兔肠源性多器官功能障碍综合征(MODS)模型复制前后的血清蛋白质组表达差异,探索该技术在家兔肠源性MODS研究中应用的可能性.取家兔肠源性MODS模型复制前后血清各50μL,去除高丰度蛋白后用丙酮将洗脱液中的蛋白沉淀,加入水化液使沉淀中的蛋白充分裂解,离心后取上清液上样.双向电泳后银染,用PDQuest7.4软件进行胶图分析,比较两组血清双向电泳图谱中的差异蛋白点.家兔肠源性MODS前后血清双向电泳图谱清晰,分辨率高,重复性好,可以满足进一步研究的需要.其中11个点在复制MODS后表达上调,8个点下调.复制家兔肠源性MODS前后血清双向电泳图谱存在显著差异;初步建立的家兔血清蛋白质双向凝胶电泳技术,将为进一步开展疾病的血清蛋白质组学研究奠定基础.
For exploring the possibility of proteomic technique's application in the study of gut originated MODS, the technique of two-dimensional polyacrylamide gel electrophoresis (2-DE) was used to compare protein expression levels in the rabbit serum which were collected before and after the model of gut originated multiple organ dysfunction syndrome (MODS). 50 μL rabbit serum which were collected before and after the model of gut originated MODS. After abundant protein depletion,proteins in the elutriant were precipitated with acetone and redissolved in rehydration sample buffer. After centrifugalization, the supernatant fluid was used as sample. After 2-DE, the gels were stained by sliver staining and analyzed by PDQuest 7.4 to compare the difference of protein expression between the two groups. In the present study, distinct two-dimensional electrophoregrams with higher resolution and better reproducibility which can be used for further investigation were obtained. Total 11 proteins were up-regulated and 8 proteins were down-regulated in gut originated MODS rabbits's serum. The results indicated that there was a significant deviation between normal rabbits and gut originated MODS rabbits in two-dimensional electrophoregrams of serum, and showed that the technique of 2-DE for rabbit serum proteome which could be effectively applied in serum proteomics of diseases.
