不育患者精液中PAF的变化及外周血PBMC上PAF-R的表达 被引量:3

摘要 目的:探讨男性不育患者精液血小板活化因子(PAF)、血小板活化因子乙酰水解酶(PAF-AH)活性的变化,以及外周血中单个核细胞(PBMC)PAF受体(PAF-R)的表达。方法:对10例男性不育患者(其中5例为精液异常组,5例为不明原因不育组)采用生物法检测精液中PAF含量,酶水解底物显色法测定PAF-AH活性,流式细胞仪检测PBMC上PAF-R的表达。并与5例有正常生育功能的男性健康体检者对照。结果:精液异常组PAF含量(1.87±0.76)μg/L,PAF-AH活性(23.56±6.86)μmol/(min.L)、PBMC上PAF-R的表达与健康体检者比较差异有非常显著性意义(P<0.01);不明原因不育组PAF含量(2.12±0.81)μg/L,PAF-AH活性(21.65±5.13)μmol/(min.L)、PBMC上PAF-R的表达与健康体检者比较差异也有非常显著性意义(P<0.01);精液异常组与不明原因不育组相比差异无显著性。结论:男性不育患者精浆PAF降低、PAF-AH升高,PAF、PAF-AH、PAF-R三者在男性不育的发生中可能起重要作用。 Objective: To study the concentration of platelet-activating factor(PAF) and the activation of PAF-Acetyl-hydrolase (PAF-AH) in sema of infertile man as well as the expression of PAF-Receptor (PAF-R) in peripheral blood PBMC of these men. Methods: Ten men with infertility( five men with abnormal semen and five men with unexplained infertility) and five men with normal fertility ability were enrolled in this study. The levels of PAF in semen were determined by the washed platelet aggregation test (bio-assay) and the activity of PAF-AH was determined by the changes of substrate assay. The expression of PAF-R on PBMC was examined by cytoflowmetry. Results: The results showed that the concentration of PAF and the activity of PAF-AH in the group with abnormal semen are significantly lower than that in control group [ ( 1.87 ± 0. 76) μg/L vs (8. 47 ±1. 52) μg/L , P 〈 0. 01 ] . The concentration of PAF and the activation of PAF-AH in the group with unexplained infertility are also significantly lower than that in control group. Furthermore, the expression of PAF-R on PBMC induced markedly in peripheral blood of men with infertility (P 〈 0.01 ). But comparing the PAF levels ,the activity of PAF-AH and the expression of PAF-R on PBMC in the men with abnormal semen and the men with unexplained infertility, there was no significant difference. Conclusion: In the men with infertility, PAF levels in semen and the expression of PAF- R on PBMC are obviously reduced, while the activity of PAF-AH is markedly increased. The changes of PAF and PAF-AH and the expression of PAF-R in PBMC are related to the male infertility.
