摘要
以来源于火炬松(PinustaedaL.)成熟合子胚的白色粘性胚性愈伤组织为材料建立了胚性细胞悬浮系,测定了其培养物的鲜重、细胞体积的增长特性及培养液中的蔗糖浓度、pH值和电导率等在培养过程中的变化规律.结果表明,在培养周期内,培养液中的蔗糖浓度、pH值和电导率的逐步降低与培养物的鲜重和细胞体积的逐步增加保持一致性.在培养的第18~21d,蔗糖浓度、pH值和电导率均接近或降到最低点,而鲜重及细胞体积的增长都达到最高点.
Embryogenic cell suspension cultures were established from white mucilaginous embryogenic calli derived from mature zygotic embryos of loblolly pine (Pinus taeda L.) Changes in the increase of fresh weight and cell packed volume in cultures, as well as pH, electrical conductivity, and sucrose concentration in medium were measured during the culture period. The results show that, in a culture circle, decrease in sucrose concentration, pH, and conductivity in medium is relative to increase in fresh weight and cell packed volume in cultures. Sucrose concentration, pH, and conductivity were the lowest, and fresh weight and cell packed volume were the highest during 18~21th day of culture
