阿霉素中毒大鼠肺组织中β-防御素-2的基因表达

摘要 目的:探讨阿霉素(adriamycin,ADM)所致大鼠急性肺损伤对肺组织β-防御素-2(BD-2)基因表达的影响。方法:雄性SD大鼠40只,随机分成ADM低剂量组(10 mg/kg)、中剂量组(20 mg/kg)、高剂量组(40 mg/kg)及对照组,每组10只。模型组分别一次性腹腔注射不同剂量ADM制备急性肺损伤模型;对照组用生理盐水腹腔注射。3天后剖胸取肺,采用半定量RT-PCR技术检测肺组织BD-2 mRNA表达的变化。结果:ADM中、高剂量组大鼠BD-2基因表达均较对照组明显上调(P<0.01),低剂量组与对照组无统计学意义(P>0.05)。结论:ADM所致大鼠急性肺损伤中,肺组织BD-2的mRNA表达水平上调。 Objective:To observe the influence of acute pulmonary injury induced by adriamyein(ADM) on β-defensin-2 (BD-2) gene expression in rats. Methods: Forty male Sprague-Dawley rats were randomly divided into four groups: low close ADM group, medium dose group,high close group and control group. The ADM groups were administered ADM of different doses ( 10,20 and 40 mg·kg^-1·d^-1). Meanwhile, normal saline was given to the control group. All the rats were killed three days later, and the lung tissues were removed by thnraeotomv. The expression of BD-2 at mRNA level was determined by RT-PCR analysis. Results:The level of BD-2 geue expression in medium-dose and high-dose groups was up-regulated significantly compared with that in the control group (P 〈0. 01 ); and the difference had no statistical significance between the low-dose group and the eontrnl (P 〉0.05 ). Conclusions:The level nf BD-2 gene is up-regulated in the lung tissues of rats with acute puhnnnary injury induced by ADM.
