摘要
采用高效液相色谱法对爬山虎红色素中原花青素的含量进行了测定。色谱柱:C18柱(日本岛津5μm;150mm×4.6mm),柱温:25℃,流动相:V(甲醇):V(水)=90:10,检测波长:280nm,进样量:20止,流速:1ml/min。结果表明在0.50-7.5μg/mL范围内具有良好的线性关系,回收率为99.96%,相对标准偏差为1.0%,原花青素的平均含量为6.8μg/mL。结论是本方法简便、快速、准确、可靠,可为爬山虎叶子提取物红色素中原花青素的含量测定的方法。
The content of oanthocyanidins in red pigment of leafage of Parthenocissus tricuspidata Planch was determined by HPLC on a Shirnadzu C 18 column (5μm; 150mm×4.6mm) with the mobile phase of 90% methanol and 10% water. The column temperature, flow rate and detection wavelength were 25℃, 1 ml/min and 280 nm, respectively. The results showed that there was a good linear relationship between absorption and proanthocyanidins content within the range of 0.50-7.5μg/mL. Recovery rate and RSD of the method were of 99.96 % and 1.0%, respectively. The average content of proanthocyanidins was 6.8μg/mL. This method is simple, fast, accurate and reliable, suitable for determination of proanthocyanidins content in red pigment from leafage of Parthenocissus tricuspidata Planch.
