清肝活血方及其拆方对酒精性肝病大鼠血浆内毒素水平及KC活化状态的影响 被引量:2

摘要 目的:探讨清肝活血方及其拆方对酒精性肝病(alcoholic liver d isease,ALD)大鼠血浆内毒素水平及KC活化状态的影响。方法:雄性W istar大鼠100只,SPF级,随机分成空白组、CC l4组各10只,余80只为造模组,采用复合因素复制ALD模型6周。空白组予等量生理盐水对照。CC l4组予微量CC l4腹腔注射2次/周。造模4周后将模型组随机分成4组,清肝活血方及其拆方组各15只,余为模型组。予等效剂量清肝活血方及拆方灌胃2周。检测血清ALT、AST水平;留取肝脏标本进行HE染色;采用偶氮显色法检测血浆内毒素水平;免疫组织化学染色检测肝组织CD68表达。结果采用SPSS12.0统计软件进行单因素方差分析及两两比较。结果:(1)清肝活血方及其拆方均可明显改善ALD大鼠肝脏脂肪变及肝脏炎症程度,清肝活血方>活血方>清肝方,清肝活血方优于清肝方。(2)清肝活血方能显著降低模型大鼠血清ALT水平,清肝方和活血方无明显作用;清肝方、活血方、清肝活血方均可明显降低ALD大鼠血清AST水平,组间比较无显著差异。(3)清肝活血方及其拆方均能降低模型大鼠血浆内毒素水平,组间比较无显著差异。(4)清肝活血方及其拆方均可抑制模型大鼠KC活化,活血方优于清肝活血方,清肝活血方优于清肝方。结论:清肝活血方及其拆方发挥对ALD的防治作用,其作用机理可能与降低血浆内毒素水平和抑制KC的活化有关。 Objective :To study the effect of Qinggan Huoxue Recipe(QGHXR) and its separated recipes on the level of plasma endotoxin and kupffer cells' active state in rats with ALD. Methods:One hundred male Wistar rats were divided into three groups randomly ,experiment group (80 rats) was intragastrieally infused mixed liquor and intraperitoneally injected CC14 twice a week,control group (10 rats) was given normal sALDne,and CC14 group (10 rats) was intraperitoneally and continuously injected CC14 twice a week. Two rats were sacrificed for histological observation per one week. Four weeks later the experiment group was divided into 4 groups randomly: QGHXR group (15 rats), Qinggan recipe group (QGR, 15 rats), H uoxue recipe group (HXR, 15 rats), the others belonged to the model group. All rats were given suitable drugs two weeks. All rats were sacrificed at the end of the sixth week. The levels of ALT,AST in the serum were analyzed. Pathological changes in liver tissues were observed in HE staining section. Plasma endotoxin levels were detected by azoic color reaction. The expression of CD6s in the liver with immunohistochemical staining. Differences between groups were evaluated by ANOVA, flowed by Fisher' s test for post hoc analysis or rank sum test. Results : ( 1 ) QGHXR and its separated recipes improved the degree of liver steatosis and inflammation, and QGHXR surpassed QGR. (2) QGHXR degraded the level of ALT in the blood of model rats,but QGR and HXR did not show manifest effect. The three recipes degraded the level of AST, and there were not significant deviation. ( 3 ) QGHXR and its separated recipes could decrease the level of endotoxin, and no.significant deviation appeared among group comparison. (4) QGHXR and its separated recipes could in- hibit the activation of kupffer cells' activation, HXR surpassed QGHXR, QGHXR surpassed QGR. Conclusion : QGHXR and its separated recipes may provide protection against ALD in rats through decreasing the level of
