绿茶多糖的扫描电镜制样新方法及原子力显微镜观察 被引量:19

摘要 利用扫描电镜(SEM)和原子力显微镜(AFM)对绿茶多糖及酶解绿茶多糖试样进行显微成像,得到颗粒状或由单个颗粒聚集成的形貌结构。结果表明:改进制样方法的扫描电镜观察结果与原子力显微镜成像基本一致,说明了改进后的扫描电镜制样方法是一种简单有效的方法。实验结果还在一定程度上解释了绿茶多糖经酶解后其微观形态的变化情况。 A new way has been found that dropping the polysaccharides on a fresh mica, fixing them with 1% O3O4 for two hours, sticking them on sample holder directly and plating them with platinum, then a clear figure of microdot structure was under the scanning electron microscope. The fine microstructure of polysaccharides distilled from green tea which was in accordance with the results from scanning electron microscope was more studied with the tipping mode atomic force microscope and show how the advanced structure reflects the change between the polysaccharides and the enzymed polysaccharides in a certain extent. So the test results told us that the new method is a simple and efficient way.
