摘要
目的 探讨烟曲霉素类似物AGM-1470抗肿瘤血管生成的作用机制及其临床应用.方法 将人结肠癌低分化细胞株SW620 CCL227移植于C57BI/6小鼠体内,18天后治疗组以AGM-1470干预.比较两组移植瘤与体积变化及肿瘤组织病理学结构.以Student t检验做统计学处理.结果 AGM-1470治疗组肿瘤体积较对照组缩小4倍余(P<0.01),治疗组平均肿瘤重量亦显著缩小(P<0.01).解剖与病理学观察表明,治疗组移植瘤表面毛细血管明显减少,新生毛细血管芽十分缺如.结论 AGM-1470具明显抗肿瘤血管生长作用,有较强肿瘤抑制作用.
PURPOSETo investigate the antiangiogenic effectiveness and potential application of fumagillin analogues AGM-1470. METHODS A single cell suspension of SW 620 (1×106/0. 1ml) was implanted to C57 BI/6 mice (n= 14) for both study and control groups. AGM-1470 were subcutaneously injected every other day to all groups of mice after the tumor were visible and palpable (150-200 mm3). The average tumor volume and weight were calculated by width2 × lenth × 0. 52 and examination of anatomy and pathology was carried out. RESULTS The average tumor volume by AGM-1470 in the study group decreased 4 times (P<0. 01) while the tumor weight also decreased 4 times (P<0. 01) in comparison with the control group. Morphological study showed that new capillary blood vessels in the AGM-1470 group were markedly inhibited. CONCLUSION AGM-1470 has markedly antiangiogenic effect and may have extensive clinical application.
