摘要
目的:研究金针菇多糖(FVP)对小鼠免疫细胞产生肿瘤坏死因子-α(TNF-α)、干扰素-γ(INF-γ)、白细胞介素-2(IL-2)的影响。方法:MTT比色法检测细胞代谢活力,ELISA方法检测TNF-α、INF-γ、IL-2的含量。结果:FVP(200、100、50μg/mL)可增强小鼠脾细胞及腹腔渗出细胞代谢MTT的活力,促进小鼠脾细胞分泌TNF-α、IFN-γ、IL-2,促进小鼠腹腔渗出细胞分泌TNF-α,并且以促进TNF-α分泌作用最为明显;FVP(100、50、25 mg/kg)可以提高荷S180瘤小鼠血清TNF-α、IFN-γ的含量。结论:FVP可能通过促进小鼠免疫细胞产生TNF-α、IFN-γ、IL-2而发挥免疫调节功能。
Objective :To study the effect of Flammulina velutipes polysaccharides (FVP) on the production of tumor necrosis factorα (TNF-α),interferonγ( INF-γ) and interleukin 2 (IL-2) by murine immunocytes. Methods:The cell's metabolic activity was determined with methylthiazolyl tetrazolium(MTT) colorimetry assay and the amounts of TNF-α ,INF-γ and IL-2 were measured by ELISA. Results:FVP (200,100,50 μg/mL)could promote the metabolic activity of murine splenocytes and peritoneal exudate cells(PEC) and increase the amounts of TNF-α, INF-γ and IL-2 in the supernatants of splenocyte cultures,and the amount of TNF-α in PEC cultures, with the most marked increase on TNF-α level. FVP( 100,50,25 mg/kg)could raise the serum levels of TNF-α and INF-γ in S180 tumor-bearing mice. Conclusion:FVP may regulate murine immune function through promoting the production of TNF-α, INF-γ and IL-2.
