摘要
目的观察大气细颗粒物(PM2.5)对培养的大鼠血管平滑肌细胞(VSMC)增殖细胞核抗原(PCNA)、血管细胞黏附分子-1(VCAM-1)表达及增殖的影响,了解其可能的作用机制。方法采集湛江市区大气中的PM2.5,以10、20、50、100、200mg/L剂量染毒培养的大鼠VSMC24h后,观察PM2.5对VSMC的PCNA、VCAM-1表达(免疫组化法)及增殖(MTT法)影响。结果在10-200mg/L范围内,随着染毒质量浓度增加,VSMC的PCNA水平从(16.67±2.58)上升到(58.00±8.25)(F=82.23,P〈0.01),VCAM-1水平从(15.83±2.32)上升到(60.83±2.56)(F=258.63,P〈0.01),MTT值从(0.49±0.067)增加到(0.72±0.025)(F=27.98,P〈0.01)。结论PM2.5促进体外培养的VSMCPCNA、VCAM-1表达,刺激VSMC增殖,在10-200mg/L质量浓度范围内具有剂量依赖性。
Objective To observe the effect of particulate matter (PM2.5) on the expression of PCNA and VCAM-1 and proliferation of rat vascular smooth muscle ceils (VSMCs) in vitro. Methods After VSMCs had been treated by PM2.5 collected from urban area of Zhanjiang for 24 hours, with doses of 10, 20, 50, 100 and 200 mg,/L, their expression of PCNA and VCAM-1 and proliferation were determined by immunohistochemical method and MTT, respectively. Results With the increase of PM2.5 concentration, expression of PCNA and VCAM-1 and MTT value in VSMCs were upregulated from (16.67 ± 2.58) to (58.00±8.25) (F=82.23,P〈0.01), (15.83±2.32) to (60.83±2.56) (F=258.63,P〈0.01), and (0.49± 0.067) to (0.72±0.025) (F=27.98,P〈0.01), respectively. Conclusion PM2.5 can in vitro induce the dose-dependent overexpression of PCNA and VCAM-1 and proliferation of VSMCs at the dose range between 10 and 200 mg/L.
