摘要
目的探讨体外培养小梁细胞表面整合素表达及其意义。方法对牛眼小梁细胞进行原代及传代培养,并行鉴定。对传三代的牛眼小梁细胞施加整合素β1一抗,后分别给予荧光和多聚辣根过氧化物酶(HRP)标记的二抗,并将细胞置于荧光倒置显微镜下观察摄像。结果应用免疫荧光和免疫酶技术方法均可观察到培养小梁细胞表面整合素β1阳性染色。结论体外培养的牛眼小梁细胞表达整合素β1,为以后研究原发性开角型青光眼病因学提供良好实验基础。
Objective To study the expression and significance of integrin-β1 in cultured bovine trabecular meshwork(BTM) cells Methods BTM cells were primarily cultured and subcuhured, and rendered identification. The third generation BTM cells were rendered rabbit Anti-Integrin- β1 , stained with anti-integrin-β1 by FITC-IgG and HRP-IgG. The BTM cells was observed by inverted micro- scope, and taked pictures. Results Making use of immunofluorescence and immunohistochemistry, we observed positive staining of integrin-β1 in cultured BIM cells. Conclusion Cultured bovine trabecular meshwork cells express integrin-β1 ,and can be used as good experimental base for researching etiology of POAG in the future.
