Cu(Ⅱ)-席夫碱配合物与DNA相互作用的电化学及荧光光谱研究 被引量:3

Studies on the Interaction Between Cu(II)-Schiff Base Complex and DNA by Electrochemistry and Fluorescence Spectroscopy

摘要 在pH 5.0的0.2 mol.L-1的NaOAc-HOAc缓冲溶液中,运用电化学和荧光光谱分析法研究了金属配合物CuL(O,O,N-2-(2,4-二羟基苯亚甲氨基)丙酸根.咪唑合铜(Ⅱ))与鲑鱼精DNA的相互作用。CuL与DNA作用后,氧化峰电流减小,式电位负移;溴化乙锭(EB)-DNA体系在加入CuL前后的荧光强度基本没有改变。研究结果表明,CuL可以通过静电作用与DNA结合,当R<2.57(R=[CuL]/[DNA]])时,化合物的化学计量系数m=0.847≈1,化学平衡常数Ka=2.41×103L.mol-1,其结构式为DNA-CuL;当R>3.14时,m=2.372≈2,其化学平衡常数Ka=3.80×109L2.mol-2,其结构式为DNA-(CuL)2。 The interaction between CuL propanoate · imidazole copper (Ⅱ))and (O, O, N-2-(2,4-dihydroxybenzylideneamino) salmon sperm DNA in 0.2 mol · L^- 1 NaOAc- HOAc buffer solution (pH = 5.0) was studied by electrochemistry and fluorescence spectroscopy. The peak current of CuL decreased with negative shift of the formal potential in the presence of DNA compared with that in the absence of DNA. The fluorescence intensity of (EB)-DNA system was unquenched before and after the addition of CuL. The results showed that CuL combined with dsDNA by electrostatic interaction. The compounds formed by the combination were experimentally examined to give rise to the stoichiometric coefficient (m), association constant (K,). The chemical formula was depicted as DNA-CuL when R was less than 2.57 (R= [-CuL]/[-DNA]) with m=0. 847 1 and Kα = 2.41 )〈 10^3 L · mol^-1. The chemical formula was depicted as DNA-(CuL)2 when R was more than 3.14 with m=2. 372 ≈2 and Kα=3.80×109 L^2· mol^-2.
