直接薄层层析技术快速检出黄曲霉毒素产生菌株 被引量:1

摘要 直接硅胶板薄层层析快速检出黄曲霉毒素产生菌株,简化了分离培养、萃取纯化等繁琐步骤,大大缩短样品检测所需时间,做到快速灵敏。检验时取生长待测菌珠的琼脂块,直接以硅胶薄层板展开浸出溶液,在短波紫外先下产毒株色谱带中毒素斑点清晰易辨。所用试剂、设备简单,费用低廉,容易掌握,无假阳性结果。尤其适用于大量菌株分离筛选。应用此方法,检测了308个真菌菌株的产毒能力。 A rapid method for search out aflatoxin. ─ producing fungi strains in foods and environment by using Thin ─layer chromatography (TLC) were reported. Preferably , the culture grown on rose-bengal agar at 30℃for at least one week. The mycelium block with agar from the centre of colony was cut out.and the extraction of the block was carried out on silica gel plates. and the qualitative determination of aflatoxin B1 and related toxins were performed with using UV analysis. Comparison with the conventional method . the advantage of TLC method was much simple and rapid. Without pseudo─positive result . 36 strains of toxin─ producing fungi were detected out of 308 strains tested using this method.
