摘要
以野生锥栗幼叶为对照,采用硅胶干燥法,设置不同保存时间(10 d,2、4、6、8、10、11、12、13、14、15、16、17、18个月),对保存不同时间的锥栗叶样抽提基因组总DNA,利用紫外分光光度计及琼脂糖凝胶电泳检测提取DNA的浓度、纯度、得率及质量等指标,评价不同保存时间对基因组DNA的影响。结果表明:随着保存时间的延长,基因组DNA的浓度呈逐渐降低趋势,12个月后,降解加剧,纯度很低。叶样硅胶保存时间在10 d至10个月之间为最佳DNA抽提时间,所得DNA浓度、得率高,纯度较好,电泳谱带清晰。
Compared with fresh samples, dried with silica gel, the genomic DNA was extracted from Castanea henryi leaves, which preserved for 10 days, 2 months, 4 months, 6 months, 8 months, 10 months, 11 months, 12 months, 13 months, 14 months, 15 months, 16 months, 17 months, 18 months, by improved CTAB methods. The concentration, purity, yield and molecular size were measured by ultraviolet spectrophotometer and agarose gel electrophoresis. The results showed that the longer the preserved time, the lower the concentration of genomic DNA. After 12 months, the degradation was serious and the purity was extremely low. It was suggested that 10 days to 10 months after stored by silica gel was the best period to extract the genomic DNA, the concentration and purity was high, the bands was good.
