摘要 目的:通过氯仿抽提法提取变异链球菌黏结素蛋白。方法:变异链球菌国际标准菌株经活化、液体培养,其表面的黏结素成分能分泌到液体培养基中,通过氯仿抽提法提取变异链球菌黏结素蛋白。对黏结素成分进行SDS-PAGE电泳分析、含量测定和抗原性检测。结果:在细菌培养液中提取到了变异链球菌表面分泌的黏结素成分。SDS-PAGE电泳分析显示是一组混合蛋白;蛋白含量测定黏结素蛋白浓度为0.7mg/ml;抗原性检测显示提取蛋白中含有变异链球菌抗原,具有抗原活性。结论:氯仿抽提法能快速提取变异链球菌黏结素蛋白。 Objective: To obtain the Streptococcus mutants by chloro-form extraction method. Methods: Streptococcus mutants strains was active and liquid cultured by international standards. In the process of liquid culture, the elements of adhesion could be secreted into culture medium. After caring on dialytic analysis, content and activity were tested by the method of chloro-form. Results: The adhesion of Streptococcus mutants was extracted by chloro form. SDS-PAGE showed that the extract was crude protein, including many kinds of protein. The content was 0. 7mg/ml. The proteins secreted through them showed activity after meeting with antibody. Conclusion: The adhesion of Streptococcus mutants can be extracted by chloro form quickly.
