摘要
以香稻1号为材料,对64对PstⅠ-MseⅠ选扩引物进行筛选,确定19对引物能扩增出适量且清晰的条带。以19对引物对具有不同表型性状的4种T5代变异株系及其对照进行AFLP分析,结果表明,变异株系基因组较香稻1号发生了明显变化。回收克隆了变异株06-5-114-11-2的p38-114特异片段和06-5-108-11-3的p82-108特异片段,并在NCBI数据库中进行序列比对,结果显示,p38-114片段与序号为NM_001074855.1的mRNA序列相似性为99%,p82-108片段与序号为NM_001061652.1的mRNA序列相似性为100%。
The rice variety Xiang 1 was used as material to screen 64 pairs of Pst Ⅰ -Mse Ⅰ selective primers,from which 19 pairs was confirmed to produce the appropriate and clear amplified bands. Then the 19 pairs of primers were used to do AFLP analysis on four T5 generation variant lines with different phenotype characters and their two parents. The result showed that the genome of variants changed significantly in comparison to Xiang 1. The specific fragments p38 -114 from variant 06 - 5 - 114 - 11 - 2 and p82 - 108 from variant 06 - 5 - 108 - 11 - 3 were cloned and sequenced,and their base similarities were compared in NCBI database,which indicated that p38- 114 presented 99% sequence similarity with mRNA NM_001074855.1 and p82 - 108 had 100% sequence similarity with mRNA NM_001061652.1.
