摘要
目的观察积雪草(CA)对2型糖尿病大鼠内皮细胞功能的影响。方法尾静脉注射链脲佐菌素(STZ)并高糖高脂饲料喂养诱导2型糖尿病大鼠模型。随机分为CA中剂量组、CA高剂量组、二甲双胍组、模型组与正常组。观察体重、口服葡萄糖耐量试验(OGTT)、三酰甘油(TG)、总胆固醇(TC)、血清胰岛素(FINS)、血管性血友病因子(vWF)的改变。结果模型组大鼠体质量明显增加(P<0.01),OGTT、TG、TC、FINS、vWF水平均较正常组明显增高(P<0.01)。与模型组比较,CA高剂量组与二甲双胍组大鼠体重均明显下降(P<0.01),且二者间无明显的统计学差异,3组治疗组大鼠餐后血糖值均明显下降(P<0.01),其中CA高剂量组与二甲双胍组之间无明显的统计学差异。与模型组比较,3组治疗组大鼠TG、TC、FINS、vWF水平均明显下降(P<0.01),其中CA中剂量组TC较二甲双胍组下降更为明显(P<0.05),CA高剂量组FINS、vWF水平较CA中剂量组(P<0.05)及二甲双胍组(P<0.01)下降更为明显。结论2型糖尿病大鼠存在糖、脂代谢紊乱及内皮细胞损伤,CA具有明显降低糖尿病大鼠血糖水平,减轻体重,降低血脂、FINS、vWF的水平,保护内皮细胞功能的作用。
Objective To study the effects of Centella Asiatica(CA) on vascular endothelial function in type 2 diabetic rats. Methods The model of type 2 diabetes mellitus in rats was established by feeding with high fat and high caloric diet and injection of streptozotocin from tail vein. Diabetic rats were randomly divided into normal group,model group,metformin group,high dosage CA group and middle dosage CA group. Weight,oral glucose tolerance test (OGTT),serum levels of triglyceride (TG) and cholesterol (TC),serum fasting insulin concentration (FINS) and VonWilly Brand factor (vWF) were detected. Results Compared with the normal group the weight,the results of OGTT,serum levels of TG,TC,FINS and vWF of rats in the model group increased significantly(P〈0.01). The weight of rats in metformin group and high dosage CA group decreased significantly compared with the model group (P〈0.01). There was no difference between rats in metformin group and high dosage CA group. Postpradial glucose levels in three treatment groups decreased remarkably (P〈0.01) and there was no difference between rats in metformin group and high dosage CA group. Compared with the model group levels of TG,TC,FINS and vWF decreased significantly (P〈0.01) and TC concentration in middle dosage CA group were much lower than that in metformin group (P〈0.05). Levels of FINS and vWF in high dosage CA group were lower than those in middle dosage CA group (P〈0.05) and metformin group (P〈0.01). Conclusion Glucose and lipid disturbance and damage of endothelial function existed in rats with type 2 diabetes. It is suggested that CA could reduce concentrations of blood glucose, plasma lipids,insulin and vWF, which shows good protective effects of relieving damage of vascular endothelium.
