摘要
目的探讨氯化三乙基锡(TETC)抗体外培养大鼠C6胶质瘤细胞增殖作用及观察C6胶质瘤细胞超微形态学变化.方法采用四甲基偶氮唑盐(MTF)比色法检测0,0.5,1.0,2.0mol/L TETC作用C6细胞24,48h增殖率.采用透射电镜观察C6细胞超微形态学变化.结果0.5,1.0,2.0mol/L TETC在体外可抑制C6胶质瘤细胞的增殖,增殖率存在着剂量依赖和时间依赖下降关系.0.5,1.0,2.0mol/L TETC作用C6细胞48h后可见细胞收缩体积变小,胞浆浓缩,细胞核内染色质趋边凝集,排列于核膜内侧,细胞核浓缩碎裂凋亡坏死的超微结构变化.结论TETC可抑制体外培养C6胶质瘤细胞增殖,其机制可能和凋亡坏死有关.
Objective To study the effects of triethyltin chloride (TETC) on proliferation and uhrastructure changes of cultured rat C6 glioma cells in vitro. Method MTF assay was performed to evaluate the effect of 0, 0.5,1.0 and 2.0 mol/L TETC on proliferation of rat C6 glioma cells for 24 h and 48 h. Electron microscope was performed to observe the ultrastructure changes of C6 glioma cells. Results 0.5,1.0 and 2.0 mol/L TETC could inhibit the proliferation of cultured C6 glioma cells in vitro performed by MTT assay and the proliferation rate decreased in a dose-dependent and time-dependent manner. The nuclear chromatic condensation and margination were found in C6 glioma cells treated by 0.5,1.0 and 2. 0 mol/L TETC for 48h by electron microscope,which showed the early ultrastructure changes of apoptosis. Condensation of cytoplasm, pyknosis, karyorrhexis, swelling of endoplasmic reticulum and mitochondria were also seen by electron microscope, which showed the ultrastructure changes of apoptosis and necrosis. Conclusion TETC can inhibit the proliferation of cultured rat C6 glioma cells in vitro, and its mechanism may be involved in apoptosis and necrosis.
