摘要
目的研究映山红总黄酮(TFR)对脑缺血及再灌注损伤的保护作用。方法采用线栓法复制局灶性脑缺血(MCAO)/再灌注损伤模型,评价动物行为功能,测定脑梗死范围,脑组织中含水量和血清中超氧化物歧化酶(SOD)、乳酸脱氢酶(LDH)的活性和丙二醛(MDA)、一氧化氮(NO)的含量。结果在局灶性脑缺血/再灌注损伤中,TFR(50.0、25.0、12.5mg/kg)连续灌胃给药5d能缩小缺血/再灌注后脑梗死体积,改善神经功能状态,降低脑含水量;减少血清中MDA、NO含量,升高SOD活性,降低LDH的活性。结论TFR对局灶性脑缺血/再灌注损伤有保护作用,其机制可能与抗自由基和脂质过氧化抑制NO的释放有关。
Objective To study the protective effect of TFR ( total flavones of rhododendra) against cerebral ischemia-reperfusion injury. Methods TFR was administrated by gavage once a day for 5 days. Focal cerebral ischemia/ reperfusion injury was caused by occluding the right middle cerebral artery (MCAO). The numerical data, which were from infarct volume, neurological deficit, brain water content, levels of malondialdehyde (MDA) , nitric oxide (NO) contents, lactate dehydrogenase (LDH) activity in plasma, levels of superoxide dismutase (SOD) activity in plasma, were evaluated. Results TFR (50. 0,25.0,12.5 mg/kg) dramatically decreased infarct volume, ameliorated the neurological deficit and reduced the brain water content. The activities of SOD in plasma were enhanced, while the activity of LDH in plasma and the contents of MDA and NO in plasma were reduced. Conclusion TFR has protective effects for cerebral injury in rats, which might be associated with its attenuating free radical and lipid peroxidation and regulating the NO release.
