体内外连续筛选建立人前列腺癌高转移细胞亚系及其生物学特性

Establishment and characterization of human prostatic carcinoma subline with high metastatic potential by selecting in vitro and in vivo successively

摘要 目的建立人前列腺癌小鼠转移模型,筛选前列腺癌高转移细胞亚系。方法采用DU145细胞悬液原位注射法接种BNX小鼠,建立前列腺癌小鼠COI模型,取肺转移灶体外培养并采用侵袭小室体外筛选高侵袭性细胞,扩增后进行原位注射,如此将肺转移灶癌细胞在小鼠体传代2次,筛选高转移细胞亚系。结果(1)原位细胞悬液注射法接种10只小鼠均成瘤,并都伴有肠系膜、腹股沟淋巴结,其中3只发现肺转移。(2)获得高转移前列腺癌细胞亚系DU145-M3,其侵袭能力强,肿瘤转移相关基因MAPK4和TIPM-2低表达,MMP-2高表达。结论COI法是有效的前列腺癌转移模型制作方法;体内外连续筛选可获得高转移细胞亚系。 Objective To establish metastatic model of haman prostate cancer cell line and to screen the subline with high metastatic potential. Methods DU145 prostate cancer cell suspension was injected orthotopically into BNX mice to construct prostate cancer cell orthotopic injection(COI) models. Metastases in the lung were cultured and then screened in transwells. The cells that penetrated the matrigel and attached to the lower-chamber compartments were harvested and expanded for the second-round selection in vivo. The same procedure was repeated twice to screen the subline with high metastatic potential. Results ( 1 ) All the BNX mice were orthotopically implanted sucessfully, taking along mesentery and groin lymph node metastasis, 3 mice had lung metastasis. ( 2 ) A subline DU145-M3 with high metastatic and invasion potential was established. The expression of prostate cancer metastatic associated gene:MAPK4,TIPM-2 was downregulated and MMP-2 was upregulated. Conclusion COI is the most effective way for prostate cancer metastatic model, and prostatic carcinoma subline with high metastatic potential can be obtained by selecting in vitro and in vivo successively.
