摘要
为了快速测定烟草及其制品中的质体色素,考察了萃取溶剂、萃取方式、萃取时间、流动相、柱温和柱流速对测定结果的影响,建立了同时测定烟草样品中6种质体色素的方法,并采用该法测定了10种烟草样品中的质体色素。结果表明:①烟草样品中的色素用90%丙酮溶液超声提取20min效果较好;②用Waters Nova-Pak-C18作色谱柱,异丙醇和乙腈-水(体积比80∶20)为流动相,梯度洗脱,流速0.5mL/min,6种色素均能完全分离;③该法的回收率为96.7%~110.7%,RSD均小于5%;④2种新鲜的冻干烟叶样品中均检测出了新黄质、紫黄质、叶黄素、叶绿素b、叶绿素a、β-胡萝卜素等色素,且其含量较高,而原烟及卷烟烟丝中仅检测出了叶黄素和β-胡萝卜素,且其含量也较新鲜烟叶低得多。该方法适合批量烟草样品中这6种质体色素的快速分析。
A reversed-phase HPLC method was developed for simultaneous quantification of 6 plastid pigments in tobacco sample. The extraction solvent, extraction mode, extraction time, mobile phase, column temperature and flow rate were tested and optimized. The plastid pigments in 10 tobacco samples were quantitatively determined, the results showed that: 1 ) the plastid pigments in tobacco were satisfactorily extracted with 90% acetone aqueous solution under ultrasonic for 20 minutes; 2) 6 plastid pigments were completely separated by reversed-phase HPLC with a Waters Nova-Pak-C18 column under a gradient elution with isopropyl and acetonitrile-water (v:v = 80: 20) as mobile phase at a flow-rate of 0.5 mL/min; 3) the recoveries of pigments were from 96.7% to 110.7% with the RSD of under 5% ; 4) 6 plastid pigments, including neoxanthin, violaxanthin, lutein, chlorophyll-b, chlorophyll- a, and β -carotene, were detectable and with relatively higher content in 2 samples of freeze-dried fresh tobacco, while only lutein and β -carotene were found in cured tobacco and cigarettes, and moreover, at far lower concentration comparing with fresh tobacco. This method is suitable for determining the 6 plastid pigments in batches of tobacco samples.
