补肾化瘀方对不孕大鼠促排卵及卵巢促黄体生成素受体表达的影响 被引量:8

Study on effect of Bushenhuayufang on promoting ovulation and Luteinizing hormone receptor expression in the ovary of sterile rat

摘要 目的:探讨补肾化瘀方对肾虚血瘀型不孕促进卵泡发育及排卵的可能机制。方法:建立雄激素致大鼠不孕模型,通过阴道图片筛选各组大鼠,采用光镜、原位杂交、免疫组织化学等方法,对补肾化瘀方治疗前后的雄激素致不孕大鼠的卵巢的形态学及其卵巢促黄体生成素受体(LHR)mRNA的表达进行观察。结果:中药组、西药组与模型组在卵巢重量及各级卵泡数目上比较差异有统计学意义(P<0.05);中药组与西药组在各级卵泡数目上比较差异有统计学意义(P<0.05)。模型组LHRmRNA的表达无论在表达面积和表达强度上都不及正常组、中药组和西药组(P<0.05);中药组与正常组在表达面积和表达强度上比较无差异。结论:补肾化瘀方可以显著增加大鼠卵巢重量及各级卵泡总数;且通过影响大鼠卵巢局部LHRmRNA的表达,促进卵泡发育,从而达到促排卵的目的。 Objective: To study the mechanism of BushenhuayuFang based on tonifying kidney and dissolving blood stasis in treating sterility with kidney deficiency and blood stasis syndrome. Methods: ARS model was set up and screened by vaginal smear. Morphological change of ovary were observed by light microscope and Luteinizing hormone receptor mRNA expression of ovary was determined by in situ hybridization and immunohistochemistry in kidney-deficiency and blood-stasis sterile rats before and after treatment. Results: There had significant difference among Chinese Medicine group, western medicine group and model group in ovarian weight and the number of follicles at all levels (P〈0.05).There was a significant difference (P〈0.05) between Chinese medicine group and western medicine group at all levels of follicle number. The LHRmRNA expression of model group was lower than normal group, Chinese medicine group and western group (P〈0.05). There was no difference between Chinese medicine group and normal group. Conclusion: Bushenhuayufang based on invigorating kidney and dissolving blood stasis can greatly increase the weight of ASR rats' ovary and the quantities of ovarian follicle of different levels in ovary. By affecting the expression of LHR mRNA in ovary, it can improve the follicular development and promote ovulation.
