大鼠供肺冷保存离体灌注模型的建立 被引量:2

摘要 目的建立以充氮膜肺、体外循环机为基础的离体肺灌注平台,用以研究移植供肺保存的效果。方法选取36只SD大鼠,用4℃LPD液经左心房逆行灌注,低温保存6 h后经离体灌注装置进行试验,经热交换器后血液温度维持在37℃,氮气与血流量比为2:1,可造成静脉血,以此灌注肺动脉,同时给予呼吸机辅助呼吸,采取经肺静脉流出道的血液行血气分析,评估供肺功能。结果整个灌注过程中各个时段流入供肺的静脉血血气分析显示PO2、PCO2无显著差异(P>0.05),可提供满意的静脉血,流出道PO2升高、PCO2降低,可反应供肺氧合能力。结论采用此灌注模型可将动脉血转化为静脉血,并持续灌注供肺,并能较好的反应供肺的功能。 Objective To establish the isolated lung perfusion model with extrocopreal circle apparatus and extracorporeal membrane oxygenation filled with nitrogen so as to study the preservation of donor long. Methods Thirty-six SD rats' donor lungs underwent perfusion with 4 Celsius degrees LPD from left ventricle and six hours isehemia, the lung arteries were perfused by the venous blood made by membrane oxygenation filled with nitrogen, and the isolated lungs were ventilated by ventilator. Gas blood exchange analysis was measured by means of sampling blood from pulmonary vein. Results The change in PO2 from pulmonary artery blood was of no difference in the whole experiment. Conclusion Simple and reliable rat model for determining the fuction of the donor lung can be provided by the means.
