摘要 为给八仙花的大规模工厂化生产提供理论基础,以八仙花当年生枝条为材料,比较研究不同激素配比条件下不同部位外植体愈伤组织的诱导及快速繁殖。结果表明,八仙花茎段、叶柄、叶块均可作为离体培养的外植体,其中叶柄的培养效果最好。在附加6-BA、NAA不同质量浓度组合的MS培养基上,均可高频诱导出愈伤组织,经再分化可获得无性系芽,继而产生丛生芽;将高度大于3 cm的增殖芽接种于1/2MS+IBA0.5 mg/L培养基上可诱导产生不定根,生根率高达100%。生根苗采用"二步法"移栽,成活率为98%。 In order to provide theoretical basis for industrialized mass production, callus induction and rapid propagation techniques were researched by media supplemented with different hormone combinations, taking different explants from branch of Hydrangea macrophylla. The results showed that stem-segment, petiole and leaf piece could be used as explants, but petiole was best. Callus could be gained with high frequent in MS medium supplemented with different 6-BA and NAA combinations. Buds generated and proliferated through callus differentiation. Buds over 3 cm height could form adventitious roots in 1/2 MS supplemented with IBA 0.5 mg/L, and the ratio of rooting was up to 100%. The survival ratio of seedlings was 98% by two-step transplanting.
