小干扰RNA对Jurkat细胞CXCR4基因表达的影响

摘要 目的研究小干扰RNA(siRNA)对Jurkat细胞趋化因子受体4(CXCR4)基因表达的影响。方法用针对CXCR4基因的siRNA转染人急性T细胞性白血病细胞系Jurkat细胞,用RT-PCR方法检测其CX-CR4基因表达水平的变化。结果CXCR4siRNA转染Jurkat细胞48h后,其表达水平下降,能明显抑制CX-CR4基因的表达。与空白对照组和阴性对照组比较,其差异有统计学意义(P<0.01)。结论利用RNA干扰技术能有效抑制靶基因的表达,为下一步有关Jurkat细胞CXCR4基因的研究奠定一定的基础。 [Objective] To study the effects of Snlall interfering RNA(siRNA) on the expression of CXCR4 in Ju-rkat ceils. [Methods] The siRNA was transfected into Acute T-lymphoblastive leukemia of Jurkat cells. The cellular CXCR4 expression was detected by RT-PCR. [ Results ] The siRNA could obviously suppression the expression of CXCR4. When transfected with siRNA for 48 h,the levels of the expression of CXCR4 were reduced (P 〈 0.01). [ Conclusion] The siRNA express vector could successfully inhibit the expression of target gene. And it can be potentially useful in the next research of CXCR4.
