摘要
目的观察沉默AnnexinⅡ(A2)基因对骨髓瘤RPMI8226细胞增殖影响。方法采用A2siRNA转染人骨髓瘤RPMI8226细胞后应用real-timePCR和流式细胞术鉴定干扰效果。MTT法检测A2siRNA对细胞增殖的作用。结果A2siRNA组较阴性对照组细胞生长抑制率在24h、48h、72h分别为16.87%±3.97%,34.92%±4.15%,39.62%±5.67%vs-8.93%±9.67%,-5.98%±2.56%,3.85%±5.48%(P〈0.05),A2siRNA可对RPMI8226细胞增殖起抑制作用。结论SiRNA沉默A2基因可抑制骨髓瘤RPMI8226细胞增殖。
Objective To investigate the effects ofAnnexinⅡ ( A2 ) gene silencing by siRNA on proliferation of multiple myeloma cell line RPMI8226. Methods A chemically synthesized siRNA duplex targeting to A2, which had been validated to be efficient in downregulation of A2 expression previously,was transfected to multiple myeloma cell line RPMI8226. Then transfection efficiency was analyzed by real-time PCR and flow cytometry.MTT assay for cell proliferation.Results Compared with those in the negative controls,the cell proliferation inhibitory rates of the RPMI8226 cells transfected in A2siRNA were significantly increased at 24h,48h, and 72h(16.87% ± 3.97%, 34.92% ± 4.15%,39.62%± 5.67% vs -8.93% ± 9.67%,-5.98% ±2.56%,3.85% ± 5.48%) (P〈0.05). It show that silencing A2 gene by siRNA had inhibited the RPMI8226 cell proliferation.Conclusion A2 gene silencing by siRNA can inhibit the proliferation of cells RPMI8226(P〈0.05).
